Job Purpose

The Oligo Synthetic Chemist (Senior Research Associate) is responsible for performing oligonucleotide synthesis, purification, and characterization as per project requirements. The role involves operating automated oligo synthesizers, troubleshooting synthesis issues, and documenting experimental results. The candidate is expected to work independently or as part of a team to ensure efficient and high-quality oligo synthesis.


Key Responsibilities

Oligo Synthesis & Purification

  • Independently operate automated oligo synthesizers (e.g., K&A-H16, Mermade-192, ÄKTA Oligosynth).
  • Synthesize DNA, RNA, antisense oligonucleotides (ASOs), and modified oligonucleotides.
  • Perform chemical conjugation using reactions such as amide coupling, click chemistry, and disulfide bond formation (S-S reaction).
  • Develop purification methods using RP-HPLC, IEX, SEC, and PAGE techniques.
  • Characterize chemically modified and conjugated oligonucleotides using analytical techniques such as LC-MS, MALDI, HRMS, and interpret the data accurately.
  • Ensure timely analysis and documentation of synthesized samples.

Instrument & Laboratory Management

  • Operate and maintain HPLC, ÄKTA-Pure, and other purification instruments.
  • Ensure proper calibration, preventive maintenance, and cleanliness of laboratory instruments.
  • Troubleshoot and resolve instrument-related issues in coordination with maintenance teams.
  • Follow MSDS guidelines and EHS protocols for safe chemical handling and waste disposal.

Documentation & Compliance

  • Record all experimental observations, results, and resources used in electronic lab notebooks (ELN).
  • Ensure strict adherence to GLP, safety, and confidentiality protocols.
  • Participate in internal audits and regulatory inspections when required.

Team Collaboration & Process Improvement

  • Plan and conduct scientific experiments in collaboration with team members.
  • Contribute to process improvements and method optimizations in oligonucleotide discovery.
  • Train and mentor junior team members in oligonucleotide synthesis and purification techniques.
  • Maintain effective communication with cross-functional teams for project execution.

Educational Qualification

  • M.Sc. in Chemistry, Biochemistry, or Biotechnology.

Technical & Functional Skills

  • Strong knowledge of solid-phase oligonucleotide synthesis (nmol to mmol scale).
  • Hands-on experience with automated oligo synthesizers and purification instruments.
  • Proficiency in purification techniques (HPLC, IEX, PAGE, SEC) and data analysis.
  • Familiarity with chemical conjugation techniques and reaction mechanisms.
  • Experience in troubleshooting synthesis and purification challenges.
  • Excellent analytical interpretation skills using LC-MS, MALDI, HRMS.
